Cyclooxygenase-1 (COX-1) is a crucial enzyme responsible for the biosynthesis of prostaglandins, which regulate various physiological functions, including gastric mucosal protection. Nonsteroidal anti-inflammatory drugs (NSAIDs) inhibit COX-1, preventing the production of prostaglandins which maintain gastric integrity by promoting mucous secretion and vasodilation. This review explores the molecular mechanism of COX-1 catalysis, detailing the oxidation of arachidonic acid into prostaglandin intermediates and the impact of NSAID inhibition on gastrointestinal health.
